Molecular Analysis of t ( l 1 ; 19 ) Breakpoints in Childhood Acute Leukemias

چکیده
MLL is fused to ENL or ELL in acute leukemias that contain t(l1; 191(q23;p131. Although ENL and ELL localize to chromosome 19, bands p13.3 and p13.1, respectively, these breakpoints are not always readily distinguished by standard cytogenetics. We therefore used reversa transcriptasepolymerase chain reaction (RT-PCR) assays to analyze 26 cases of childhood acute leukemia containing t(l1; 19) to determine the frequencies of ENL and ELL involvement. All 17 cases of acute lymphoblastic leukemia (ALL) had MLLIENL

متن کاملConsistent involvement of the bcr gene by 9;22 breakpoints in pediatric acute leukemias.
To investigate the relationship of bcr-abl fusion mRNAs with childhood acute lymphoblastic leukemias (ALL), we examined 27 pediatric Philadelphia chromosome (Ph1)-positive acute leukemias using a reverse polymerase chain reaction (PCR) procedure. In cells from 24 leukemias, single bcr-abl PCR products were detected that corresponded to breakpoints in the minor breakpoint cluster region (mbcr in...
متن کاملThe Chromosome 4 q 21 Gene ( AF - 4 / FEL ) Is Widely Expressed in Normal Tissues and Shows Breakpoint Diversity in t ( 4 ; 1 l ) ( q 21 ; q 23 ) Acute Leukemia
The chromosomal translocation, t (4 ; l l )(q21 ;q23), is the most common type of 1 1 q23 chromosomal abnormality, being highly prevalent in infant acute leukemias and associated with a poor prognosis. The t (4 ; l l ) results in the fusion of an ll q23 gene (MLL, HRX, Htrx-1 , or ALL-1) and a 4q21 gene (AF-4 or FEL). To further evaluate the 4q21 gene and its role in t(4;ll) acute leukemia, we ...
متن کاملRAPID COMMUNICATION Predictability of the t(1;19)(q23; p13) From Surface Antigen Phenotype: Implications for Screening Cases of Childhood Acute Lymphoblastic Leukemia for Molecular Analysis: A Pediatric Oncology Group Study
The t(l ;I 9)(q23;pl3) translocation occurs in approximately 5% of 6-precursor acute lymphoblastic leukemias (ALLs) occurring in children. Its presence has been associated with a poor prognosis, which may be overcome with more intensive therapy. Although leukemic cells from cases of t(1; 19)-ALL frequently express cytoplasmic p heavy chains, their complete antigenic profile remains undefined. A...
متن کاملRAPID COMMUNICATION ENL, the Gene Fused With HRX in t ( l1; 19) Leukemias, Encodes a Nuclear Protein With Transcriptional Activation Potential in Lymphoid and Myeloid Cells
Chromosome band 1 lq23 is the site of recurring translocations with a variety of partner chromosomes in myeloid and lymphoid acute leukemias, infant leukemias, and treatmentinduced secondary acute myelogenous leukemia. The translocation breakpoints cluster in a restricted region of the HRX gene resulting in fusion genes that encode an N-terminal portion of Hrx fused to various partner proteins....
